Induction of triploidy with caffeine treatment in the African catfish (Clarias gariepinus)

چکیده مقاله:
Induction of triploidy is one of the biotechnological methods in aquaculture used for genetic manipulation. It refers to a state where organisms have three complete sets of chromosomes instead of two and can result in sterility. Caffeine treatment that is safe and inexpensive, serve to induce triploidy in catfish. To suppress the second meiotic division, fertilized eggs were exposed to three different concentrations (5, 10 and 15 mM) of caffeine solution for 20 min beginning at 3 min after fertilization. After that, the eggs were incubated at ambient temperature until hatching. The induction of triploidy in fry was determined for three concentrations of caffeine by means of flow-cytometric analysis. The lowest rate of triploidy (20.40 ± 1.13%) was obtained in the group treated with 5 mM caffeine and the highest (69.10 ± 2.18%) in the group treated with 15 mM caffeine. Our results suggest that caffeine can be used to induce triploidy in catfish.

Histopathological Effects of Glyphosate on Juvenile African Catfish (Clarias gariepinus)
Most aquatic herbicides have undergone some toxicity testing for effects on non-target aquatic organisms, little of this testing has been conducted on early life stages of Clarias gariepinus. African catfish C. gariepinus were exposed to acute concentrations of glyphosate for 96hrs. The lethal concentration (LC ) 5O value of glyphosate was 0.295 mg l for 96h of exposure. The mean mortality perc...

In order to estimate responses of growth rate and growth hormone of Clarias gariepinus to stocking density, fish (initial weight 30.71 ± 0.89 g) were stocked in 120-L tanks at densities of 35, 65, 95, and 125 kg/m3 for 60 days. On day 30 and 60, the fish growth rates were measured, pituitary growth hormone (GH) mRNA levels and serum GH concentrations were examined by real-time qPCR and ELISA te...

متن کاملHistomorphological Assessment of Spermatogenesis in African Catfish Clarias gariepinus with reference to Spermatogonial Proliferation
The fresh water catfish is the most predominant species of the teleost bred in captivity in Nigeria and has provided a model for the study of reproduction in aquatic vertebrates, but information on the number of mitotic divisions and spermatogonial generations remains inconclusive. This investigation aimed at assessing the number of generations of spermatogonia produced prior to meiotic divisio...
متن کاملOntogeny of Swimming Movements in the Catfish Clarias gariepinus
The swimming movements of C. gariepinus larvae were recorded with a high-speed camera (400, 500 and 800 fps) from 0 to 336 hours post-hatching. Movements of adult fish were also recorded to provide information on the last developmental stage. Seven landmarks positioned on the fish midline were used during tail beating to determine various parameters during ontogeny and, on the basis of these pa...
